But now, scientists and wellness enthusiasts alike are turning their attention to the tiniest components of our cells and exploring their potential link to how well, and perhaps how long, we live.

NAD+ (nicotinamide adenine dinucleotide) and its precursor NMN (nicotinamide mononucleotide) are two compounds that have become a growing focus of research into the biology of ageing. But what exactly are they, how do they work and why are researchers so interested in them? With insight from Dr Laure Verstraeten, we’ll explore more about these tiny molecules and their potential.

What’s NAD+?

Life as we know it simply can’t exist without this tiny molecule, as it’s found in all living cells. NAD+ is a coenzyme, which means it helps enzymes (proteins which drive chemical reactions) within your body to do their jobs efficiently.1 Without coenzymes like NAD+, these enzymes can’t work properly and some bodily processes can slow down. One of NAD+’s most important roles is in the mitochondria, which are where most of your cells’ energy is generated. Here, NAD+ helps convert nutrients from food into energy. NAD+ also supports processes that help your cells repair everyday wear and tear, communicate properly and keep your energy and metabolism running smoothly.2

However, “as we age, NAD+ levels are thought to decline, which has been linked to changes in cellular processes involved in energy metabolism and repair,” explains Dr Verstraeten.3 This has led scientists to explore NAD+ biology in greater detail, including the roles of compounds such as NMN in NAD+ production.

What’s NMN?

If NAD+ is the engine that powers your cells, then NMN is one of the key components that helps keep that engine running. Think of it as a building block, or a precursor, meaning the body uses it to make NAD+ through a series of chemical reactions.4

Without precursors like NMN, the body’s ability to make enough NAD+ can be limited – and potentially even more so as we get older, according to early research.4 Like your NAD+ levels, your NMN levels may also decline as you age, perhaps due to the body not producing as much while using more.5

NMN is one of the more direct ways your body can replenish its NAD+ levels, making it a key focus of interest in longevity research. But NAD+ doesn’t rely solely on NMN; it has other precursors too, like nicotinamide (NAM, a form of vitamin B3), nicotinic acid (NA) and nicotinamide riboside (NR).2

Why are scientists interested in NMN and NAD+?

“The decline of NAD+ as we age has captured the attention of research groups around the world because it appears to play a role in many aspects of health,” Dr Verstraeten explains. “Numerous animal studies suggest that when NAD+ levels decline, mitochondria (the cell’s energy factories) may work less efficiently, DNA repair processes can be impaired and cells may become more vulnerable to stress. In these models, NMN supplementation raises NAD+ levels and appears to improve several of these age-related cellular functions.”6

This has led scientists to ask the question: could maintaining NMN and NAD+ levels in humans also help cells stay resilient for longer and add healthier years to our lives?

Could NAD+ and NMN supplements help with ageing?

While interest and research in this area have grown rapidly, especially around how these molecules might influence biological ageing, current human evidence is still emerging.7 Scientists are still learning how they work in the human body over the long term, who may benefit most and how they fit into overall health.

What we do know from the emerging evidence is that NMN supplementation can lead to an increase in NAD+ levels in adults, with some studies suggesting improvements in aspects of general health.8,9 It’s also clear that these molecules play a meaningful role in the body’s energy systems and that alone makes them an intriguing area of research (especially when we talk about ageing well rather than simply living longer).10

However, Dr Verstraeten tells us, “while these early findings are promising, we do need more research. So, while NMN and NAD+ may play a supportive role in healthy ageing, more high-quality human studies are needed before wider claims can be made.”6

Can you get NAD+ from food?

We all know that eating a varied, balanced diet with lots of fruits and vegetables is best for our health and wellbeing, and turns out, this may also help support our NAD+ levels. Dr Verstraeten explains that, “while NAD+ itself isn't directly obtained from the diet, it can be formed in the body from several precursors found in food, including NA and NAM (vitamin B3 forms), tryptophan and NR.”11 

Good sources of these include nuts, legumes, grains, poultry and dairy, the pillars of a balanced and varied diet. “NMN is found in many plants but in small quantities and it’s unclear whether dietary intake alone can meaningfully raise NAD+ levels. That’s why supplementation with NAD+ precursors has been studied more closely.”12

How to support your body as you age

Whilst the jury may still be out on whether NAD+ and NMN support healthy ageing, the good news is there are plenty of evidence-based ways to support your body as you get older - starting with everyday habits, like:
  • eating a healthy, balanced diet: incorporate plenty of fruits, vegetables, lean protein and whole foods (like the Mediterranean diet), while limiting ultra-processed foods like sugary drinks, processed meats and fast food13
  • moving regularly: aim for 150 minutes per week of moderate physical activity like walking or cycling, or ramp it up and aim for 75 minutes of intense activity, like running or aerobics – whatever gets your heart pumping14 
  • keeping your brain engaged: reading, doing puzzles, learning a new skill or playing games can all help keep your brain active15,16
  • drinking up: stay hydrated by drinking plenty of water every day – try to aim for around eight glasses or 2-2.5 litres throughout your day17
  • connecting with others: combat loneliness and isolation by keeping in touch with friends, family and engaging with your community18,19
  • quitting smoking: research shows that quitting smoking, no matter how long you’ve been smoking for, can improve your health20
  • limiting your alcohol consumption: drinking too much alcohol can affect your health in so many ways, from your energy levels to your weight and even your skin21
  • getting some good sleep: practice healthy sleep hygiene and aim for eight hours of good quality sleep every night22,23
  • managing your stress: find healthy stress management techniques that work for you to help keep stress at bay24,25
  • staying on top of your health: make sure to attend regular health screenings and check-ups, as these can help manage illnesses as well as monitor aspects of your health like your blood pressure and cholesterol levels26,27

The final say

When it comes to healthy ageing, it’s easy to hope for a miracle supplement that holds all the answers. NAD+ and NMN have certainly sparked excitement, with the science suggesting they play important roles in how our cells age and repair themselves, but the story is still unfolding. Ageing is a complex journey we all take, shaped by how we move, eat, sleep and so much more.
